Large scale HPLC in process or production scale is usually a costly operation! Large volumes of solvents are pumped through the column, and expensive equipment and packing is used. In addition, the product is often very valuable, and must be recovered in as large quantities as possible. To optimize the chromatography so that the chromatography is run as efficiently as possible is a very difficult task, due to the complex relations between the parameters involved.
At Eka Chemicals we realized this early on, and to be able to assist companies with developing efficient purification methods we developed an optimization program, KromaGuide.
The KromaGuide program is run as a free service for our customers, where there is a large application. For development work it is normally not justified to perform an optimization, since time, not the cost involved, is the critical factor.
KromaGuide will give you the best running parameters, like optimal flow rate, amount loaded, column length etc, and you will also obtain a sensitivity analysis that tells you how the costs will increase if you deviate from the optimal conditions. The cost breakdown in system, solvent, packing, labor and crude cost is also presented.
To perform an optimization we need only 3-4 experiments run at analytical conditions, and at least 1, preferably 2-3, overloaded experiments, run with different loading amounts. We will then analyze the data with the help of KromaGuide, to obtain the best running conditions for your application.
